Here's just a few examples of what I mean.
1) Fortify base.
Requires that you clear a POI, and once clear build fortifications in specific points (doors, spikes, torches, etc) to prepare a POI for use by other people.
2) Lure Zombies
Requires that you enter a POI and place demolition charges in certain spots along with a 'zombie lure'. Once placed after a certain amount of time the charges go off (Whether or not the player is present!) and the POI gets trashed.
3) Salvage Run
Spawns 3-5 special blocks in a POI that must be salvaged with a wrench/ratchet/impact driver for a unique resource that's turned in to complete the quest. Salvaging these blocks makes noise that draws extra zombies to you.
4) Clean up POI.
Someone dumped a bunch of contaminated materials in a POI, and it's mutating zombies that wander in. The material needs to be cleared out and stored to prevent more mutations.
This mission would spawn a bunch of materials that have to be deposited into a special container in the POI, carrying them slowly builds up a debuff that infects you when it reaches a certain amount of stacks.
Now obviously it wouldn't have to be exactly these, but I really do think that added variety would be wonderful.
